Four amino-acid substitutions (D-Ala², Gln⁸, Ala¹⁵, Leu²⁷) protect it against DPP-4 cleavage and trypsin digestion while preserving full GHRH receptor affinity.

The Drug Affinity Complex (DAC) is a maleimide group that binds serum albumin and stretches the half-life to roughly 6–8 days, producing a sustained "bleed" of growth hormone. Without DAC, the half-life is around 30 minutes — which keeps GH release sharply pulsatile and much closer to natural secretory physiology. That makes this form the standard choice for research into pulse amplitude rather than total GH exposure.
