What Is SS-31?
SS-31, also known as Elamipretide or MTP-131, is a water-soluble, cell-permeable tetrapeptide (D-Arg-2',6'-dimethyl-Tyr-Lys-Phe-NH₂) belonging to the Szeto-Schiller class. Its alternating aromatic-cationic motif allows it to cross cell membranes without a transporter and concentrate several thousand-fold on the inner mitochondrial membrane, where it binds the phospholipid cardiolipin.
Key Mechanisms
- Cardiolipin binding — stabilises cristae architecture and the cardiolipin–cytochrome c interaction
- Electron-transport efficiency — preserves supercomplex assembly, improving ATP output per unit oxygen
- ROS reduction — limits electron leak at complexes I and III, lowering superoxide generation at source
- Peroxidase suppression — prevents cardiolipin from converting cytochrome c into a peroxidase, an early apoptotic trigger
